Yohimbine

Stimulantoral · inferred

Indole alkaloid and alpha-2 adrenergic receptor antagonist. Purified yohimbine is not interchangeable with yohimbe bark: analyses of retail supplements have found large and sometimes unlabeled variation in yohimbine content. Parent-drug exposure varies markedly with CYP2D6 genotype and inhibitors, so a single curve is only a population estimate. Sympathetic effects can include anxiety, tremor, increased blood pressure, and tachycardia; regulatory status varies by country.

Research behind this entry

  1. Biopharmaceutics and metabolism of yohimbine in humans Le Corre P, Dollo G, Chevanne F, Le Verge R · European Journal of Pharmaceutical Sciences, 1999 DOI

Healthy volunteers received 5 mg intravenously and 8 mg orally. Mean absolute bioavailability was 22.3% but highly variable, renal excretion of unchanged parent was about 0.1%, and active 11-hydroxy-yohimbine had a…

  2. Oral Yohimbine as a New Probe Drug to Predict CYP2D6 Activity Vay M et al. · Clinical Pharmacokinetics, 2020 DOI

In 16 genotype-characterized healthy volunteers, clearance after 5 mg varied from 25.3 to 15,864 mL/min. Paroxetine reduced clearance more than fivefold in extensive metabolizers but not poor metabolizers, confirming…

  3. Yohimbine as a treatment for erectile dysfunction Wibowo DNSA, Soebadi DM, Soebadi MA · Turkish Journal of Urology, 2021 DOI

This systematic review and meta-analysis included eight randomized trials. Pooled results favored yohimbine-containing interventions, but the monotherapy subgroup did not significantly improve the broader sexual…

  4. Pharmaceutical quantities of yohimbine found in dietary supplements in the USA Cohen PA, Wang YH, Maller G, DeSouza R, Khan IA · Drug Testing and Analysis, 2016 DOI

Laboratory analysis of 49 retail products found 0 to 12.1 mg yohimbine per recommended serving. Only 11 labels stated a specific amount, most stated amounts were inaccurate, and only two products combined an accurate…
